DNase degradation assay. Plasmid DNA (pEGFP-N1) was digested with ECORI and NOTI and then incubated with DNase-I at 37°C with increasing concentrations of MgCl2 or CaCl2 overnight. First lane, Molecular marker; second lane: Restriction fragments of plasmid DNA and 0.05 U/μg DNase in 5 mM MgCl2; third lane, restriction fragments of plasmid DNA and 0.05 U/μg DNase in 1.67 mM MgCl2; fourth lane, restriction fragments of plasmid DNA and 0.05 U/μg DNase in 0.56 mM MgCl2; fifth lane restriction fragments of plasmid DNA and 0.05 U/μg DNase in 0.19 mM MgCl2; sixth lane, restriction fragments of plasmid DNA and 0.05 U/μg DNase in 0.06 mM MgCl2; Lane 7, restriction fragments of plasmid DNA and 0.05 U/μg DNase in 5.0 mM CaCl2; lane 8, restriction fragments of plasmid DNA and 0.05 U/μg DNase in 1.67 mM CaCl2; lane9, restriction fragments of plasmid DNA and 0.05 U/μg DNase in 0.56 mM CaCl2; lane 10, restriction fragments of plasmid DNA and 0.05 U/μg DNase in 0.19 mM CaCl2; lane 11, restriction fragments of plasmid DNA and 0.05 U/μg DNase in 0.06 mM CaCl2.
