Flow cytometry analysis with 3E8 mAb. - U937, THP-1, HL60, and MM1R cells (2 × 10⁶ cells per sample in PBS) were incubated with 3E8 mAb or goat anti-B7-H4 polyclonal antibody for 30 min at 4°C. Normal mouse IgM was used as an antibody control. Cells were washed and resuspended in a solution of goat antimouse IgM coupled with FITC or donkey anti-goat IgG coupled with FITC. Finally, cells were washed twice and samples were analyzed using a FACScan flow cytometer.
